The folic acid and L-Methylfolate for Depression

The role of folic acid in neurotransmitter (chemical messenger) synthesis and homocysteine metabolism has prompted people to study its potential impact on depression.

A cohort study in 2017 assessed the relationship between the duration of folic acid supplementation during pregnancy and the incidence of postpartum depression (PPD). Among 1592 participants, depression was assessed 6-12 weeks postnatally. People who took folic acid for more than six months had a lower prevalence of PPD than those who took folic acid for less than six months.

In addition, a systematic review and meta-analysis in 2018 evaluated the use of folic acid in the treatment of major depression. The researchers found that treatment with 5 mg or less of folic acid or 15 mg of methyl folic acid per day in combination with selective serotonin reuptake inhibitor (SSRI) was significantly beneficial compared with placebo.

On the other hand, other studies have not found any benefits. For example, a longitudinal study in 2015 assessed the impact of folic acid and other B vitamins on the risk of depression.
Participants without depression at the start of the study were evaluated for three years. Although B6 and B12 were associated with a lower incidence of depression, they were not associated with folic acid.
